Vol.4 No.2 – 1 : Protective Effects of α-lipoic acid on Biological changes Induced by α-cypermethrin in Testis Rats

Abstract

α-cypermethrin is one of the most potent insecticide used worldwide.This study was planned to evaluate the possible role of α-lipoic acid in α-cypermethrin induced toxicity in rats. The treated groups were;the control, α-cypermethrin, α-lipoic acid and α-cypermethrin and α-lipoic acid groups. Our results showed that administration of α-cypermethrin caused significant decrease in RBC count, PCV and Hb content and an increase of WBC count. Also, α- cypermethrin caused significant increase in the levels of cholesterol, TGs, LDL-Cand VLDL-C, while the HDL-C was decreased.In addition, α-cypermethrin caused reduction in serum testosterone, FSH, and LH levels in intoxicated rats. Furthermore, the co-administration of α-lipoic acid mitigated the toxicity of α-cypermethrin by partially normalizing these biochemical parameters. Our results were supported by histopathological observations of testis. Our data suggest that α-lipoic acid may have a protective role against α-cypermethrin induced toxicity in rats.

Vol.2 No.07 -9 : Molecular and cytochemical comparative assessment between the two food additives,sunset yellow and curcumin-induce testicular toxicity in mice.

Abstract

The present work was planned to study the effects of E110 (sunset yellow) as a common synthetic in Egypt and E110 (curcumin) as a natural food-drug colorants on the testis of the male mouse. The plan of work was designed to cover six parameters: histopathological, cytochemical (involving DNA and total proteins), testis weight, sperm parameters (i.e., sperm abnormalities and sperm motility), and measuring testosterone levels in blood sera. The mice were divided into three groups, ten per each. The first group remained as controls, whilst the second orally given sunset yellow-E110 (30 mg/kg b.wt/day) as SY-group and the third one E100 ‘CU-group’ also gavage 37 mg/kg b.wt., both fed on their acceptable daily intake (ADI) dosages for 60 days. The results detected that SY revealed distinct alterations in the desired parameters, particularly histological changes in structure of seminiferous tubules such as vacuolation, necrosis and multinucleate cells. Whilst, the cytochemical DNA and proteinic profiles of the SY-treatment mice exhibited severe damage in the DNA and total protein configurations. However, such deteriorations in the spermatogenic epithelia were also approved with changes in the other criteria after administration with E110. From such alterations, the E110 recorded a highly significant increase (P< 0.0001) in the abnormalities of sperm morphology and motility. Moreover, the testosterone levels in sera of male mice indicated the significant differences among groups. The molecular protocol manifested SY (E110) - induced DNA polymorphic changes in confrontation with control by primer OPC07, whilst CU (E100) kept on the control pattern. In conclusion, the present study explored the possibility of using the applied six parameters to assessment and differentiate between the two food flavours indicating that E100 (CU) is more biosafe than the synthetic additive E110 (SY).

Vol.2 No.5 -4 : Age-Related Changes in the Histological Structure of Rabbit Testes and the Prophylactic Role of Vitamin E.

Abstract

The present investigation is planned to compare between the histological structure of rabbit testes of different ages and the prophylactic role of vitamin E on senescent animals. Male NewZeland rabbits were divided into four groups according to age. Group I: represented the young rabbits (one month age, weighing 1 ± 0.4 kg), group II: adult rabbits (6 months age, weighing 4 ± 0.5 kg), group III: aged animals (24 month age, weighing 7.5 ± 0.5 kg and group (IV) aged rabbits treated daily with the therapeutic dose of vitamin E orally at a dose of 10 mg/kg b.w / day for 60 days. The testes of young rabbits revealed a small form of seminiferous tubules with regular outline with spermatogonia and Sertoli cells, and the lumen appeared empty from spermatozoa. The interstitium is made up of loose connective tissue with large polyhedral interstitial Leydig cells. In adult studied animals, testes demonstrated with all spermatogenic cells and the lumen filled with spermatozoa. In aged rabbits, the histological studies of testis demonstrated atrophied irregular tubules, disarrangement of vacuolated spermatogenic cells, and sloughing of germ cells into the tubular lumen with reduction and degeneration of spermatozoa. The interstitium between the tubules was widen and filled with degenerated and reduced Leydig cells. Administration of vitamin E to aged animals group revealed an obvious improvement of spermatogenic cells, and the spermatozoa appeared in the lumen of many seminiferous tubules.

Abstract

Ultrastructure biomarker reflects the effects of pollutants. The present study amid to evaluate the reproductive changes of the aquatic hemipteran insect Anisops sardeus (as a bioindicator organism), inhabiting three sites in Al-Mahmoudia canal (Abou-Hommous, Zarcon town, Manshia) which varied in physical and chemical properties. Mahmoudia canal is considered the main water source for Alexandria, which receive water from Rosetta branch at Mahmoudia city. The canal receives domestic and agriculture wastes from Zarcon Drain and other non-point sources. The present work is concerned with monitoring bioaccumulation of metal in the testes of A. sardeus using SEM-X-ray microanalysis and illustrating spermatogenesis disruptions. Insects caught from polluted sites (Zarcon town, Manshia) showed higher proportion of heavy metals in particular Cu, Zn and Hg than in the less polluted site (Abou-Hommous). Many alterations of the general architecture of the testis were pronounced. Disruption and damage for the normal cellular organization were observed. In epithelial cells, aggregated clumps of heterochromatin, irregular nuclear envelope, cytoplasm with disorganized mitochondria and convolution of follicular wall were noticed. In spermatogonia the nucleus appeared with disintegrated nucleolus, vacuolated cytoplasm and degenerative changes in the mitochondria. According to the obtained results the water quality of Al-Mahmoudia canal was lower at the polluted sites and the watercourse from south to north direction has been increased in pollution sources. The results also showed that the intensity of the histopathological changes increased with increasing the intensity of heavy metals. As a biomarker of exposure to toxicants, histopathology represents a useful tool to assess the degree of pollution

Abstract

This work aims to study the histopathological and histochemical changes in the testes of male albino rats post exposure to mobile phone radiation and the possible protective role of vitamin E and/or silymarin. The current experiment was carried out on 30 adult male albino rats (Sprague dawly). Rats were randomly and equally divided into five groups: 1) Group C: control rats left without treatment; 2) Group R: rats exposed to frequency equals 950 MHz of electromagnetic field (EMF); 3) Group R + E: rats received vitamin E before exposure to frequency equals 950 MHz of EMF; 4) Group R + S: rats received silymarin before exposure to frequency equals 950 MHz of EMF and 5) Group R + E+ S: rats received vitamin E parallel to silymarin before exposure to frequency equals 950 MHz of EMF. Electromagnetic field exposed rats showed testicular alterations which were ameliorated by using either vitamin E or silymarin alone. However, both vitamin E and silymarin administration ameliorated the damaging effects of testes of mobile phone exposed rats more than that of silymarin or vitamin E alone.
